Product Specific Information

The ICRF44 monoclonal antibody reacts with human CD11b, a 165 kDa adhesion molecule associated with integrin beta2 (CD18). It is expressed on monocytes, granulocytes, activated lymphocytes, and a subset of NK cells. CD11b acts as a receptor for intercellular adhesion molecules (CD54, CD102, CD50) and iC3b, playing a crucial role in cell-cell and cell-matrix interactions.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated for flow cytometry on normal human peripheral blood cells. Recommended use: 5 µL (0.4 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

Important Notes:

• NovaFluor dyes are incompatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains with NovaFluor dyes.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) must be used for optimal staining. Add 5 µL per sample before combining with cells.
• For single-color controls, use 5 µL of CellBlox Plus Blocking Buffer per 100 µL cell sample (10³–10⁸ cells).

Target Information

CD11b (integrin alpha-M, ITGAM, integrin alpha-X, ITGAX) is a 165 kDa adhesion molecule forming a heterodimer with CD18. Known as integrin alpha-M beta-2, Mac-1, or CR3, it is expressed on:

• Monocytes/macrophages
• Granulocytes
• Activated lymphocytes
• NK cells
• Dendritic cells
• Microglia

CD11b/CD18 acts as a receptor for ICAM-1, ICAM-2, ICAM-4, CD14, CD50, CD23, heparin, iC3b, fibrinogen, and Factor X. Expression varies by cell type and tissue, being higher in peritoneal macrophages than splenic macrophages. It is a common marker for microglia and monocyte maturation.

Diseases associated with CD11b dysfunction include systemic lupus erythematosus and ITGAM-related susceptibility to autoimmune disorders.
