The monoclonal antibody eBio12A5 recognizes FR4 (Folate receptor 4, FR delta, FBP3), a 35 kD glycosylated receptor for folic acid and N5-methyltetrahydrofolate.
Natural Treg cells express high levels of FR4 and, together with CD25, can be used to distinguish T cell subsets:

• FR4^high CD25^high: Natural Treg (high Foxp3)
• FR4^high CD25^low: Central memory T cells (produce IL-2 upon stimulation)
• FR4^low CD25^high: Effector memory T cells (secrete IFN-γ, IL-17)

Co-staining studies show that epitopes recognized by eBioTH6 and eBio12A5 are different, enabling functional comparison.

Applications Reported: Flow cytometric analysis
Applications Tested: Mouse splenocytes (≤ 0.5 µg/test; final volume 100 µL; 10⁵–10⁸ cells/test).
Careful titration recommended for optimal performance.

Fixation: Store samples in IC Fixation Buffer (Product #00-8222) or 1-step Fix/Lyse Solution (Product #00-5333) for up to 3 days at 4°C in the dark.

Target Information

FR4 is a glycosylated 35 kD receptor for folic acid and N5-methyltetrahydrofolate.
Tregs express high FR4 and, together with CD25, help distinguish Treg, effector, memory-like, and naive T cells.
In vivo studies with eBioTH6 show reduced CD4⁺CD25⁺ cells and enhanced tumor response, leading to elimination of advanced tumors.
