Product Specific Information

The UT41 monoclonal antibody was generated against BaF3 cells overexpressing mouse TLR4.
Toll-like receptors (TLRs) are type I transmembrane proteins with extracellular leucine-rich repeats and cytoplasmic domains homologous to the type I IL-1 receptor. Among them, TLR2 and TLR4 are pattern recognition receptors involved in innate immunity and inflammation.

TLR4 associates with MD-2 and CD14 to form a complex responsible for LPS recognition and signaling. TLR4 (~100 kDa) is expressed by myeloid lineage cells. The MTS510 antibody preferentially reacts with TLR4 associated with MD-2, while UT41 binds TLR4 regardless of MD-2 complex formation.

Applications Reported: Flow cytometric analysis.
Applications Tested: Tested on mouse thioglycolate-elicited peritoneal exudate cells. Recommended ≤0.5 µg/test (100 µL volume, 10⁵–10⁸ cells/test). Optimize concentration for best performance.

Fluorochrome Information:
Super Bright 436 is excited by the violet laser (405 nm) and emits at 436 nm.
Use a 450/50 bandpass filter or equivalent. Ensure instrument compatibility.

When using multiple Super Bright dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to reduce non-specific interactions.

Target Information

Target: TLR4 (CD284)
TLR4 is a member of the Toll-like receptor family, key in pathogen recognition and innate immunity activation. It is expressed in placenta and myelomonocytic leukocytes, responding to lipopolysaccharides (LPS) of Gram-negative bacteria.

TLR4 forms part of the LPS receptor complex with CD14 and LY96, mediating signal transduction and cytokine production. Mutations in TLR4 affect LPS responsiveness. Multiple transcript variants exist, though protein-coding potential varies.
TLR4 is expressed in peripheral blood monocytes, certain B-cells, and placenta. TLR4-deficient mice show reduced LPS response and increased susceptibility to Gram-negative sepsis.
