Product Specific Information
The GK1.5 monoclonal antibody reacts with the mouse CD4 molecule, a 55 kDa cell surface receptor expressed by most thymocytes, subsets of mature T cells, and dendritic cells. CD4 binds to MHC class II on antigen-presenting cells and plays a key role in T cell development and function. In T cells, CD4 associates with protein tyrosine kinase p56lck through its cytoplasmic tail. Binding of GK1.5 is blocked by RM4-5.
Each product includes:
- 1 vial of NovaFluor conjugate
- 1 vial of CellBlox Plus Blocking Buffer
Applications Reported: Flow cytometric analysis
Applications Tested: Tested on mouse splenocytes. Recommended ≤0.2 µg/test (in 100 µL final volume). Optimize antibody titration for best performance.
Important Notes:
- NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
- Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for compatibility.
- CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining with NovaFluor conjugates.
- Add 5 µL of CellBlox Plus Blocking Buffer per sample before combining with cells.

Target Information
The CD4 antigen is involved in the recognition of MHC class II molecules and functions as a co-receptor for HIV. It is primarily expressed on T helper cells but also found on monocytes, macrophages, and dendritic cells. CD4 expression occurs in thymus, lymph nodes, tonsils, spleen, and certain non-lymphoid tissues. It plays a critical role in T-cell activation via association with the T-cell receptor complex and Lck kinase. Multiple alternatively spliced transcripts have been identified (RefSeq, July 2017).
