Product Specific Information

The GK1.5 monoclonal antibody reacts with the mouse CD4 molecule, a 55 kDa cell surface receptor expressed by thymocytes, mature T cell subsets, and dendritic cells. CD4 binds to MHC class II molecules on antigen-presenting cells, playing a key role in T cell development and activation. In T cells, CD4 associates with protein tyrosine kinase p56lck through its cytoplasmic tail. Binding of GK1.5 is blocked by RM4-5.

Applications Reported: Flow cytometric analysis
Applications Tested: Flow cytometric analysis of mouse splenocytes (≤ 0.2 µg/test).
A test is defined as the amount of antibody for staining a sample in 100 µL volume. Cell number can range from 10⁵ to 10⁸ cells/test. Adjust titration for optimal performance.

Compatibility Note:
NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assessment.

CellBlox Plus Blocking Buffer Usage:

• Required for optimal staining with NovaFluor conjugates.
• Add 5 µL per sample before combining with cells.
• For single-color controls: 5 µL per 100 µL of cell sample (10³–10⁸ cells).

Target Information

CD4 antigen is a co-receptor for MHC class II molecules and HIV. It is primarily expressed in T helper cells and other immune cells such as monocytes, macrophages, and dendritic cells. CD4 is detectable in thymus, lymph nodes, tonsils, spleen, and certain non-lymphoid tissues. It plays a role in T-cell activation and may mediate neuronal damage in immune-related CNS diseases.
Multiple alternatively spliced transcripts have been identified (RefSeq, July 2017).
