Product Specific Information

The GK1.5 monoclonal antibody reacts with the mouse CD4 molecule, a 55 kDa cell surface receptor expressed by thymocytes, subsets of mature T cells, and dendritic cells. CD4 binds to MHC class II on antigen-presenting cells and plays an important role in T cell development and function. In T cells, CD4 associates with protein tyrosine kinase p56lck through its cytoplasmic tail. Binding of GK1.5 is blocked by RM4-5.

Each product contains:

Applications Reported: Flow cytometric analysis
Applications Tested: Mouse splenocytes (≤0.2 µg/test, 100 µL volume, 10⁵–10⁸ cells/test)

Note: NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI). Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.

CellBlox Plus Blocking Buffer (Cat. No. C001T06F01):

• Improved formulation for optimal NovaFluor staining
• Add 5 µL per sample (regardless of number of NovaFluors)
• For single-color controls: use 5 µL per 100 µL of cell sample containing 10³–10⁸ cells

Target Information

CD4 antigen is involved in recognition of MHC class II molecules and acts as a co-receptor for HIV. It is expressed primarily on T helper cells, as well as monocytes, macrophages, and dendritic cells. CD4 expression is found in thymus, lymph nodes, tonsils, spleen, and various non-lymphoid tissues. It contributes to T-cell activation via association with the T-cell receptor complex and Lck kinase. Alternative splicing variants are known (RefSeq, July 2017).
