Product Specific Information
The GK1.5 monoclonal antibody reacts with the mouse CD4 molecule, a 55 kDa cell surface receptor expressed by most thymocytes, a subset of mature T cells, and dendritic cells. CD4 binds to MHC class II on antigen-presenting cells and is crucial for T cell development and function. Binding of GK1.5 is blocked by RM4-5.
Each product includes:
- 1 vial of NovaFluor conjugate
- 1 vial of CellBlox Plus Blocking Buffer
Applications Reported: Flow cytometric analysis.
Applications Tested: Tested by flow cytometric analysis of mouse splenocytes. Recommended ≤0.2 µg/test (100 µL sample; 10⁵–10⁸ cells/test). Optimize titration for best results.
Important Notes:
- NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
- Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability analysis.
- CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
- Add 5 µL per sample to antibody cocktails/master mixes before combining with cells.
- For single-color controls: 5 µL per 100 µL of cell sample (10³–10⁸ cells).
NovaFluor Conjugates:
- Based on Phiton™ technology using novel nucleic acid dye structures.
- Designed for engineered fluorescent signatures minimizing spillover and spread.
- Excitation: 509 nm; Emission: 614 nm; Laser: 488 nm (Blue).
Target Information
CD4 antigen is involved in the recognition of MHC class II molecules and serves as a co-receptor for HIV. It is expressed primarily on T helper cells, as well as monocytes, macrophages, and dendritic cells. CD4 expression is observed in thymus, lymph nodes, tonsils, spleen, and some non-lymphoid tissues. It plays a key role in T-cell activation via interaction with Lck kinase and may mediate neuronal damage in immune-related CNS diseases. Multiple alternatively spliced transcripts exist (RefSeq, July 2017).
