This 2POK monoclonal antibody reacts with mouse ThPOK, also known as c-Krox and Zbtb7b.
ThPOK is a 78 kDa transcription factor expressed in thymocytes and T lymphocytes in spleen and lymph nodes.
In non-hematopoietic cells, ThPOK is detected in skin, heart, smooth muscle, and liver.

ThPOK promotes CD4+ lineage differentiation by inhibiting CD8 lineage-specific genes such as Runx3, and prevents Cd4 silencing during CD4+ T-cell development.
It also plays a role in the effector functions of invariant natural killer T (iNKT) cells and gamma delta T cells.

Applications Reported:

• Intracellular staining followed by flow cytometric analysis.

Applications Tested:

• Tested by intracellular staining and flow cytometric analysis of mouse thymocytes using the Foxp3/Transcription Factor Staining Buffer Set (Product # 00-5523-00).
• Use ≤ 0.25 µg/test (100 µL final volume).
• Cell number: 10⁵–10⁸ cells/test (empirically determined).
• Antibody titration recommended for optimal performance.
