This 1TMYS monoclonal antibody reacts with mouse Thymocyte-expressed molecule involved in selection (Themis), also known as Grb2-associating protein (Gasp).
Themis is a cytoplasmic protein essential for positive selection and the development of single-positive thymocytes.
Highest expression is found in CD4+CD8+ double-positive thymocytes, with lower levels in mature T cells.
Themis associates with PLCγ1 and LAT and is rapidly phosphorylated by SLP-76 following TCR signaling.

There are five isoforms of mouse Themis generated by alternative splicing. The 1TMYS antibody recognizes isoforms 1–4 but does not recognize isoform 5.

Applications Reported: Intracellular staining followed by flow cytometric analysis.
Applications Tested: Tested using intracellular Fixation & Permeabilization Buffer Set (Product #88-8824-00) according to BestProtocols®: Protocol A (two-step for cytoplasmic proteins).
Use ≤ 0.25 µg per test (100 µL final volume). Optimal cell number: 10⁵–10⁸ cells/test. Antibody titration is recommended for best performance.
