Product Specific Information

This R6H1 monoclonal antibody reacts with CD196 (CCR6), a seven transmembrane G protein-coupled receptor expressed on T, B, dendritic, natural killer, and Langerhans cells. CCR6 binds MIP-3a/CCL20, a chemoattractant for dendritic cells, effector/memory T cells, and B cells. It is involved in host defense and epithelial inflammation, and plays roles in Th17 differentiation and CD4+FoxP3+ regulatory T cell development.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and validated for flow cytometric analysis of normal human peripheral blood cells. Recommended usage: 5 µL (1.0 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

Fluorophore Information:
Brilliant Ultra Violet™ 737 (BUV737) emits at 732 nm and is suitable for instruments with a 355 nm UV laser. Ensure cytometer compatibility.

Buffer Recommendations:
When using multiple polymer dye-conjugated antibodies (Super Bright, Brilliant Violet, Brilliant Ultra Violet), use Super Bright Complete Staining Buffer (#SB-4401-42) or Brilliant Stain Buffer™ (#00-4409-75) to minimize non-specific interactions.

Light Sensitivity: Protect vial and stained samples from light.
Fixation: Samples can be stored in IC Fixation Buffer (#00-8222-49) or 1-step Fix/Lyse Solution (#00-5333-54) for up to 3 days at 4°C in dark conditions.
Methanol Compatibility: Compatible with short-term methanol fixation (<1 hour).

Target Information

CCR6 is a beta chemokine receptor predicted to be a seven transmembrane G protein-coupled receptor. It is preferentially expressed by immature dendritic and memory T cells. Its ligand, macrophage inflammatory protein 3 alpha (MIP-3α), is key for B-cell maturation and antigen-driven differentiation. CCR6 may regulate dendritic and T cell migration during immune and inflammatory responses. Alternative splicing variants encoding the same protein have been described.
