Specificity Comments:
This antibody recognizes a protein of 65–76 kDa, identified as antichymotrypsin (AACT). AACT is a plasma protease inhibitor synthesized in the liver as a single glycopeptide chain. In humans, normal serum levels of AACT are about one-tenth of plasma concentrations, which increase in response to trauma, surgery, and infection. Elevated levels are often reported in cerebrospinal fluid and plasma of Alzheimer’s disease (AD) patients. PSA and its SDS-stable complex with AACT are used as diagnostic markers for prostate cancer. AACT deficiency may contribute to chronic liver disease. This antibody reacts with histiocytes and histiocytic neoplasms and is widely used to identify histiocytes and related tumors. Acinar tumors of the pancreas and salivary gland may also show AACT positivity.

Target Information

SERPINA3 encodes Alpha 1-Antichymotrypsin (Alpha 1-ACT), an early-stage acute-phase plasma protein and serpin that preferentially inactivates chymotrypsin, cathepsin G, and chymase. Alpha-1-ACT is associated with amyloid plaques in Alzheimer’s disease and in aged human and primate brains. Regulation of serine proteases and their inhibitors is important in neuromuscular differentiation. Prostate-specific antigen (PSA), a chymotrypsin-like serine protease, is predominantly complexed to Alpha-1-ACT.
