Product Specific Information

Description:
eBioTY/11.8 recognizes CD73, a 69-kDa GPI-anchored cell-surface protein with ecto-5’-nucleotidase activity. Expression on myeloid cells (CD11b) is restricted to the bone marrow. In humans, CD73 can be induced to secrete a soluble form with IL-2, suggesting a role in activation signaling.
CD73 expression increases during lymphocyte maturation and is found on regulatory T cells as well as memory CD4 T cells (Thpp).

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Tested:

• Tested by flow cytometric analysis of mouse splenocytes
• Recommended ≤ 0.2 µg/test (in 100 µL final volume, 10⁵–10⁸ cells/test)
• Titrate for optimal performance

NovaFluor Dyes Compatibility:

• Not compatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI)
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for best results

CellBlox Plus Blocking Buffer (Cat. No. C001T06F01):

• Required for optimal staining with NovaFluor conjugates
• Add 5 µL per sample before combining with cells
• For single-color controls: 5 µL per 100 µL cell sample (10³–10⁸ cells)

Target Information

CD73 (Ecto-5’-nucleotidase):
Catalyzes conversion of purine 5’-mononucleotides to nucleosides, preferring AMP as substrate.
CD73 is a dimer of two 70 kDa subunits anchored to the plasma membrane via GPI linkage and serves as a lymphocyte differentiation marker.
Deficiency is associated with immunodeficiency diseases and calcification disorders.
Two transcript variants encoding different isoforms have been identified.
