Product Specific Information
This CXNFT monoclonal antibody reacts to mouse NOS2 (inducible NOS, iNOS). Nitric oxide synthase enzymes catalyze the formation of nitric oxide from L-arginine through an NADPH- and oxygen-dependent mechanism.
There are three isoforms of NOS: NOS1 (nNOS), NOS3 (eNOS), and NOS2 (iNOS). NOS2 is induced by bacterial endotoxins and inflammatory cytokines such as IFN-γ and TNF-α. It is expressed by M1 macrophages and myeloid-derived suppressor cells but not by M2 macrophages.
Applications Reported: Intracellular staining followed by flow cytometric analysis.
Applications Tested: Tested on mouse thioglycolate-elicited peritoneal exudate cells using the Intracellular Fixation & Permeabilization Buffer Set (Product # 88-8824-00). Recommended ≤ 0.125 µg per test.
Brilliant Ultra Violet™ 615 (BUV615) emits at 615 nm and is suitable for UV (355 nm) laser-equipped cytometers.
Use Super Bright Complete Staining Buffer (Product # SB-4401-42) or Brilliant Stain Buffer™ (Product # 00-4409-75) to minimize non-specific polymer interactions.
Light Sensitivity: Protect from light.
Fixation: Samples can be stored in IC Fixation Buffer (Product # 00-8222-49) or 1-step Fix/Lyse Solution (Product # 00-5333-54) for up to 3 days at 4°C in the dark.

Target Information
iNOS (Inducible Nitric Oxide Synthase) produces nitric oxide (NO) from L-arginine, mediating vasorelaxation, neurotransmission, and cytotoxicity.
iNOS is inducible by cytokines (IFN-γ, TNF, IL-1, IL-2) and lipopolysaccharides (LPS). It is expressed in macrophages, hepatocytes, synoviocytes, and smooth muscle cells.
Three iNOS pseudogenes are located on chromosome 17 (Smith-Magenis syndrome region).
Diseases associated with iNOS dysfunction include achalasia and impotence.
