Product Specific Information

The OKT3 monoclonal antibody reacts with an epitope on the epsilon-subunit within the human CD3 complex. It exhibits potent immunosuppressive properties in vivo and has been effective in treating renal, heart, and liver allograft rejection.
CD3 is expressed by thymocytes and all mature T cells, playing a critical role in TCR assembly and signaling.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and tested on normal human peripheral blood cells (5 µL/test).
Cell number can range from 10⁵–10⁸ cells/test.

Fluorophore Information:
PE-eFluor 610 is excited by 488–561 nm lasers and emits at 607 nm. Recommended filter: 610/20 band pass (PE-Texas Red equivalent).
Light sensitive — protect from photo-oxidation.

Fixation:
Samples may be stored in IC Fixation Buffer (Product #00-8222) or 1-step Fix/Lyse Solution (Product #00-5333) for up to 3 days at 4°C in the dark.

Target Information

The CD3 complex is essential for transducing antigen-recognition signals and regulating TCR surface expression.
CD3 subunits (gamma, delta, epsilon, zeta) are encoded on chromosome 11 and are members of the immunoglobulin superfamily.
CD3 is expressed on 68–82% of peripheral blood lymphocytes and 65–85% of thymocytes, but not on B or NK cells.
Defects in CD3 genes are associated with CD3 immunodeficiency.
