Product Specific Information

Description:
The UCHT1 monoclonal antibody reacts with human CD3e, a 20 kDa subunit of the TCR complex. The CD3 epsilon chain, along with gamma and delta subunits, is required for proper assembly, trafficking, and surface expression of the TCR complex. CD3 is expressed by thymocytes and all mature T cells. Crosslinking of TCR via immobilized UCHT1 induces intracellular activation and proliferation pathways.

Applications Reported:
This antibody is reported for use in flow cytometric analysis.

Applications Tested:
Pre-titrated and validated by flow cytometric analysis of normal human peripheral blood cells. Recommended usage: 5 µL (0.25 µg) per test (100 µL total volume). Cell number range: 10⁵–10⁸ cells/test.

Fluorophore Information:
PE-eFluor® 610 is excited by 488–561 nm lasers and emits at 607 nm. Recommended filter: 610/20 band pass (PE-Texas Red® equivalent). Ensure instrument compatibility.

Light Sensitivity:
Sensitive to photo-induced oxidation; protect from light.

Fixation:
Samples can be stored up to 3 days at 4°C in the dark using IC Fixation Buffer (Product #00-8222) or 1-step Fix/Lyse Solution (Product #00-5333).

Target Information

The CD3 complex is essential for transducing antigen-recognition signals into T cells and regulating TCR expression. It includes CD3 gamma, delta, epsilon, and zeta subunits, encoded by genes on chromosome 11. These subunits contain cytoplasmic tails with tyrosine-based motifs involved in signal transduction. CD3 antigen is found on 68–82% of peripheral blood lymphocytes and 65–85% of thymocytes, but not on B or NK cells. Defects in CD3 genes are associated with immunodeficiency.
