Product Specific Information

The 6D4 monoclonal antibody reacts with human MHC class I chain-related proteins MICA and MICB. These proteins share 83% amino acid similarity and structural homology with classical HLA molecules but do not bind beta2 microglobulin or peptides typical of HLA class I.

MICA and MICB are expressed on endothelial cells, fibroblasts, gastric epithelium, and PHA-stimulated T cells. They act as ligands for NKG2D expressed on NK cells, γδ T cells, and αβ CD8+ T cells. MICA and MICB expression has been detected in epithelial tumors from breast, lung, ovary, prostate, colon, and kidney tissues.

Applications Reported: Flow cytometric analysis, immunoprecipitation, and immunohistochemical staining.
Applications Tested: Tested by flow cytometric analysis of HeLa cells at ≤0.25 µg/test (100 µL final volume, 10⁵–10⁸ cells/test). Optimal titration recommended.

Storage and Handling: Use in a sterile environment.
Filtration: 0.2 µm post-manufacturing filtered.
Purity: >90% (SDS-PAGE).

Target Information

MICA encodes a highly polymorphic MHC (HLA) class I chain-related gene A. The protein is expressed on the cell surface but does not associate with beta-2-microglobulin. MICA functions as a stress-induced antigen recognized by intestinal epithelial γδ T cells.
