Product Specific Information

  • For immunohistochemistry (PFA fixed): incubate antibody for 30 minutes at room temperature.
  • Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 minutes at 95°C, followed by cooling at room temperature for 20 minutes.

Target Information

CD137 ligand (CD137L) is a type II membrane protein and a member of the TNF superfamily. It acts as a co-stimulatory molecule expressed on antigen-presenting cells (DCs, monocytes/macrophages, B cells) and is upregulated upon activation.
Its receptor, CD137 (4-1BB), is expressed on various cell types including inflamed endothelial cells and activated T cells. Interaction between CD137 and CD137L enhances T cell proliferation, IL-2 secretion, survival, and cytotoxic activity.
CD137L expression is observed in most B-cell lymphomas (mantle cell, follicular, diffuse large B-cell) but absent in Hodgkin and T-cell lymphomas. It is also found in carcinoma cell lines, suggesting a role in T cell–tumor cell interactions.
