The Applied Biosystems™ Human ACTB (beta actin) Endogenous Control (VIC™/TAMRA™ Probe, Primer Limited) is designed as an endogenous control for relative gene expression quantification in cDNA samples. It can be used with other TaqMan™ Gene Expression Assays and is suitable for both multiplex and singleplex PCR reactions. The probe is labeled with VIC™ and TAMRA™ dyes, and the primers are limited.

TaqMan™ Endogenous Controls simplify the selection of appropriate controls for quantifying gene expression across human, mouse, rat, and other eukaryotic genes.

Simple to Use

All components are QC-tested and formulated into a single 20X mix. They are fully compatible with universal conditions for two-step RT-PCR. Use with TaqMan™ Universal PCR Master Mix (with or without AmpErase™ UNG) and your cDNA sample to generate sensitive and reproducible data on Applied Biosystems instruments (e.g., 7900HT, 7300, 7500, 7000, 7700 systems).

Flexible Offering

Each endogenous control uses 5′ nuclease chemistry with options for different reporter dyes and quenchers:

• FAM™ dye-labeled TaqMan™ MGB probe (250 nM, final) with two unlabeled primers (900 nM each)
• VIC™ dye-labeled TaqMan™ MGB probe (250 nM, final) with two unlabeled primers (150 nM each, primer limited)
• VIC™ dye-labeled TAMRA™ probe (250 nM, final) with two unlabeled primers (150 nM each, primer limited)

Choosing the Right Endogenous Control

Endogenous controls normalize expression levels of target genes by correcting for cDNA loading differences. For best results, ensure consistent expression across all samples. For multiplexing, the control gene should be more abundantly expressed than the target.

Multiplex vs. Singleplex PCR

• VIC™-labeled probes are primer limited and suitable for multiplexing.
• FAM™-labeled probes are not primer limited and are intended for singleplex use.
