Target Information
Tryptophan hydroxylase (TPH) catalyzes the 5-hydroxylation of tryptophan, initiating the biosynthesis of serotonin and melatonin.
In mammals:
- Serotonin is synthesized mainly in neurons from the Raphe nuclei.
- Melatonin synthesis occurs in the pineal gland.
TPH activity is rate-limiting for serotonin but not melatonin biosynthesis.
The enzyme’s activity is enhanced by phosphorylation via PKA and Ca²⁺/calmodulin kinase II (CAM K II), both acting on Ser58 within the regulatory domain of TPH.
