Product Specific Information
The monoclonal antibody CXCR3-173 recognizes mouse CD183 (CXCR3), a seven-transmembrane G-protein-coupled chemokine receptor that binds CXCL9 (Mig), CXCL10 (IP-10), and CXCL11 (ITAC). CD183 plays a critical role in CD4 T cell responses to grafts, and CXCR3 knockout mice show compromised allograft rejection.
Expression is found on NK cells, subsets of T lymphocytes and Tregs, with preferential expression on Th1-polarized cells.
The CXCR3-173 antibody affects chemotaxis in response to ligand; ligand presence eliminates staining. In vivo addition delays cardiac and pancreatic islet allograft rejection.

This gene encodes a G protein-coupled receptor selective for chemokines IP10, Mig, and I-TAC. These chemokines belong to the CXC subfamily, characterized by a single amino acid separating the first two conserved cysteines. Binding induces leukocyte activation, cytoskeletal rearrangement, and chemotactic migration.
Inhibition by Bordetella pertussis toxin suggests coupling to Gi-class G proteins. Signal transduction likely involves pathways common to other chemokine receptors. Chemokine-induced desensitization occurs via phosphorylation-dependent receptor internalization, with subsequent recycling restoring responsiveness.
CXCR3 is prominently expressed in effector/memory T cells and inflamed tissues, where its ligands are locally produced. It is a key target for small-molecule antagonist development for inflammatory disease therapy. Multiple transcript variants exist, encoding different isoforms.
