Target Information
SIRP alpha (CD172a, signal-regulatory protein alpha)는 수지상세포(DC), 대식세포, 호중구 등 골수계 세포에서 발현되는 수용체형 막단백질입니다.
이 단백질은 여러 활성화된 티로신 키나아제(EGFR, PDGFR, src, insulin receptor 등)의 기질로 작용하며, 수용체 티로신 키나아제 신호전달 경로의 음성 조절에 관여합니다.
SIRP alpha는 CD47과 결합하여 세포 내 ITIM(면역수용체 티로신 억제 모티프)의 인산화를 유도하고, SHP-1 및 SHP-2 인산화효소를 활성화시킵니다.
이 상호작용은 대식세포의 식세포작용 억제, T 세포 및 DC의 양방향 활성화 등 다양한 조절 과정에 관여합니다.
또한 SIRP alpha는 혈청, 성장인자, 인슐린, 온코진, 성장호르몬 및 세포 부착에 의해 유도되는 세포 반응을 조절하며, 다양한 생리적·병리적 과정에서 중요한 역할을 합니다.
암세포는 CD47을 과발현하여 SIRP alpha를 활성화함으로써 대식세포에 의한 파괴를 회피합니다.
