Product Specific Information

The monoclonal antibody 1H9 recognizes both mouse and human Pax5 (BSAP, B cell specific activator protein), a transcription factor of the paired box (pax) family. Pax5 is the only member expressed in hematopoietic cells and plays a key role in B cell lineage commitment and differentiation. Its expression is maintained through most B cell stages and suppressed during plasma cell differentiation.

Pax5 influences heavy chain recombination and regulates expression of various B and non-B cell proteins. It is associated with several neoplasms including diffuse large B cell lymphoma (DLBCL), non-Hodgkin lymphoma, and B-cell ALL. Translocations with elastin, IGH, ETV6, FOXP1, and EVI3 have been reported.

Recommended Buffers: For optimal intracellular staining and flow cytometry, use Foxp3/Transcription Factor Staining Buffer Set

Applications Reported: Intracellular staining followed by flow cytometric analysis.
Applications Tested: Tested in normal human peripheral blood cells using the Foxp3/Transcription Factor Staining Buffer Set. Recommended ≤0.25 µg/test in 100 µL final volume (10⁵–10⁸ cells/test). Optimize titration for best results.

Target Information

PAX5 is a transcription factor of the PAX family, crucial in early development and B-cell differentiation. It acts as a B-cell lineage-specific activator protein (BSAP), expressed during early but not late stages of B-cell maturation. PAX5 expression is also found in the developing CNS and testis, suggesting roles in neural development and spermatogenesis. Mutations in PAX5 are associated with acute lymphoblastic leukemia.
