Description:
Monoclonal antibody BCL-UP recognizes Bcl-6 (B-cell lymphoma 6), a POZ family transcriptional repressor. Bcl-6 is abundantly expressed in germinal center B cells and activated B cells. This 79 kDa protein is crucial for germinal center B cell proliferation and survival, and promotes pre-B cell survival during development. It is constitutively expressed in diffuse large B cell lymphomas (DLBCLs), Burkitt’s lymphoma, and non-Hodgkin’s lymphoma. Expression is also observed in CD4+ T follicular helper and memory T cells. Bcl-6 is antagonized by Blimp-1.

Applications Reported:

• Intracellular staining followed by flow cytometric analysis.

Applications Tested:

• Tested in human tonsil cells using Foxp3 Fixation/Permeabilization Buffers (Product # 00-5521-00).
• Refer to BestProtocols® for detailed staining procedures (Protocol B).
• Recommended usage: 5 µL (0.125 µg) per test (approx. 10⁵–10⁸ cells/test).

Target Information

Bcl-6 proto-oncogene product is a Kruppel-type zinc-finger protein expressed in normal germinal center B cells and related lymphomas. It is involved in chromosome rearrangements at 3q27 in non-Hodgkin’s lymphomas, detected in 33–45% of diffuse large B cell lymphomas. Bcl-6 expression is confirmed immunohistochemically in follicular lymphoma, diffuse large B cell lymphoma, Burkitt’s lymphoma, and nodular lymphocyte predominant Hodgkin’s disease.
