Target Information
HMGB1 (High-mobility group box-1) 단백질은 원래 핵 내 비히스톤 DNA 결합 단백질로 알려졌습니다. 그러나 최근 연구에 따르면 손상되거나 괴사된 세포는 HMGB1을 세포외로 방출하며, 이는 염증성 사이토카인으로 작용합니다.
Mouse HMGB1은 215개의 아미노산으로 구성된 단일 사슬 폴리펩타이드이며, 두 개의 DNA 결합 도메인(HMG box A, aa 9–79; box B, aa 89–162)과 음전하를 띤 30 aa의 C-말단 산성 꼬리 영역을 포함합니다.
Box B는 사이토카인 활성을 가지며, Box A는 전사인자와의 상호작용에 관여합니다. HMGB1은 염증 매개체로 작용하여 단핵구 이동과 사이토카인 분비를 유도하고, T 세포–수지상세포 상호작용을 매개합니다.
또한 RAGE, TLR2, TLR4 수용체를 통해 신호를 전달하며, 거의 모든 세포의 핵과 세포질에 존재합니다. HMGB1은 무균적 염증 반응 및 감염 관련 염증 반응에서 중요한 매개체로 작용하며, 외상, 패혈증, 관절염, 허혈-재관류 손상 등 다양한 병리적 상태에서 그 수준이 상승하는 것으로 보고되었습니다.
