Product Specific Information
The GL1 monoclonal antibody reacts with mouse CD86, an ~80 kDa surface receptor also known as B7-2. CD86 and CD80 are members of the B7 family of costimulatory molecules. CD86 is expressed at low levels on B cells, macrophages, and dendritic cells, and is upregulated upon stimulation through BCR complex, CD40, and cytokine receptors. It is also expressed by activated mouse T cells and thioglycolate-elicited peritoneal cells.
CD86 interacts with T cell surface molecules CD28 and CD152 (CTLA-4), playing a critical role in T-B crosstalk, T cell costimulation, and antibody production. The upregulation kinetics of CD86 support its major contribution during the primary immune response.
Applications Reported: Flow cytometric analysis.
Applications Tested: Tested on stimulated mouse splenocytes at ≤0.125 µg/test (100 µL volume). Optimal titration recommended.
Excitation: 488 nm Emission: 520 nm Laser: Blue Laser
Filtration: 0.2 µm post-manufacturing filtered
Target Information
CD86 is one of two ligands (the other being CD80) for CTLA4 and CD28. It acts as a costimulatory molecule essential for T-cell activation during antigen presentation. Insufficient co-stimulation can induce immune tolerance. CD86 is a type I membrane protein of the immunoglobulin superfamily, expressed on antigen-presenting cells. It binds CD28 and CTLA-4 on T cells, respectively promoting or inhibiting T-cell activation. Alternative splicing produces multiple isoforms. CD86 dysfunction is associated with diseases such as gallbladder squamous cell carcinoma and myocarditis.
