Target Information

The DCTN1 gene encodes the largest subunit of dynactin, a macromolecular complex of 10 subunits (22–150 kDa). Dynactin binds to both microtubules and cytoplasmic dynein and plays roles in:

  • ER-to-Golgi transport
  • Lysosome and endosome movement
  • Spindle formation and chromosome movement
  • Nuclear positioning and axonogenesis

This subunit interacts with the dynein intermediate chain and binds microtubules via a conserved CAP-Gly domain in its N-terminus.
Alternative splicing produces multiple isoforms.
Mutations in this gene are associated with distal hereditary motor neuronopathy type VIIB (HMN7B), also known as distal spinal and bulbar muscular atrophy (dSBMA).
