Description:
The BTLA antibody (clone 6F7) reacts with mouse BTLA (B and T lymphocyte attenuator). BTLA is expressed by peripheral lymphocytes, splenic macrophages, developing B cells in the bone marrow, developing T cells in the thymus, and mature (but not immature) bone marrow-derived dendritic cells. BTLA functions as a negative regulator of activation and/or function in various hematopoietic cell types. It is reported to bind B7-H4, though further validation is needed.

Note:
The anti-mouse BTLA monoclonal antibody 6F7 stains CD4⁺ and CD8⁺ single-positive thymocytes (Hurchla et al). Other clones (8F4, 3F9.D12, 6G3, 6H6) do not stain SP thymocytes but show similar staining patterns in splenocytes and bone marrow cells. The discrepancy in thymocyte staining remains unclear.

Applications Reported:
This 6F7 antibody is reported for use in flow cytometric analysis.

Applications Tested:
Tested by flow cytometric analysis of mouse splenocytes. Recommended ≤0.125 µg/test (100 µL final volume). Cell number should be empirically determined (10⁵–10⁸ cells/test). Optimal dilution should be titrated for each assay.

Filtration:
0.2 µm post-manufacturing filtered.

Target Information

BTLA (B and T-lymphocyte attenuator):
A co-inhibitory receptor of the CD28 superfamily that functions as a lymphocyte inhibitory receptor, regulating immune responses. Constitutively expressed on most CD4⁺ and CD8⁺ T cells, its expression decreases upon activation but remains on Th1 cells (not Th2). BTLA interacts with HVEM (herpesvirus entry mediator), playing a key role in lymphocyte activation and immune homeostasis.
