Immunohistochemistry (PFA fixed): Incubate antibody for 30 minutes at room temperature.
Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 minutes at 95°C, followed by cooling at room temperature for 20 minutes.

Target Information

CD63 (LAMP-3, lysosome-associated membrane protein-3) is a glycoprotein of the tetraspanin family, present in late endosomes, lysosomes, and secretory vesicles of various cell types. It is also found in the plasma membrane following cell activation and is widely used as a basophil activation marker.
CD63 interacts with integrins, influences phagocytosis and cell migration, and is involved in H/K-ATPase trafficking and ROMK1 channel regulation. It also functions as a T-cell costimulation molecule.
Expression of CD63 can serve as a prognostic marker in early-stage carcinomas. CD63 is expressed on activated platelets, monocytes, and macrophages, and weakly on granulocytes, B, and T cells. It is identical to the melanoma-associated antigen ME491 and the platelet antigen PTLGP40.
Diseases associated with CD63 dysfunction include melanoma and Hermansky-Pudlak Syndrome.
