• Immunohistochemistry (PFA fixed): Incubate antibody for 30 min at room temperature (RT).
• Staining of formalin-fixed tissues requires heating sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 min at 95°C, followed by cooling at RT for 20 min.

Target Information

CD63 (LAMP-3, lysosome-associated membrane protein-3) is a glycoprotein of the tetraspanin family found in late endosomes, lysosomes, and secretory vesicles of various cell types. It is also present on the plasma membrane after cell activation and serves as a basophil activation marker.
CD63 interacts with integrins, influences phagocytosis and cell migration, and is involved in H/K-ATPase trafficking and ROMK1 channel regulation. It also acts as a T-cell costimulation molecule. Expression of CD63 can predict carcinoma prognosis.
CD63 is expressed on activated platelets, monocytes, macrophages, and weakly on granulocytes, B, and T cells. It is identical to melanoma-associated antigen ME491 and platelet antigen PTLGP40.
Diseases associated with CD63 dysfunction include melanoma and Hermansky-Pudlak Syndrome.
