• For immunohistochemistry (PFA fixed): incubate antibody for 30 minutes at room temperature.
• Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 minutes at 95°C, followed by cooling at room temperature for 20 minutes.

Target Information

CD63 (LAMP-3, lysosome-associated membrane protein-3) is a glycoprotein of the tetraspanin family present in late endosomes, lysosomes, and secretory vesicles of various cell types. It is also found on the plasma membrane following cell activation, making it a widely used basophil activation marker.
CD63 interacts with integrins, affects phagocytosis and cell migration, and is involved in H/K-ATPase trafficking and regulation of ROMK1 channels. It also serves as a T-cell costimulation molecule. Expression of CD63 can be used for predicting prognosis in early-stage carcinomas.
CD63 is expressed on activated platelets, monocytes, and macrophages, and weakly on granulocytes, B, and T cells. It is identical to the melanoma-associated antigen ME491 and the platelet antigen PTLGP40.
Diseases associated with CD63 dysfunction include melanoma and Hermansky-Pudlak Syndrome.
