Description:
The PC61.5 antibody reacts with mouse CD25, the 55 kDa interleukin-2 receptor alpha chain (IL-2Rα). CD25 is expressed by early progenitors of T and B cells and by activated mature T and B lymphocytes. CD25 alone binds IL-2 with low affinity but forms a high-affinity receptor when associated with CD122 (IL-2Rβ) and CD132 (common γ chain). The PC61.5 antibody inhibits IL-2 binding to both receptor classes. CD25 is involved in lymphocyte differentiation and activation/proliferation.

Applications Reported: Flow cytometric analysis.

Applications Tested:
Tested by flow cytometry using mouse splenocytes and Con A-stimulated splenocyte cultures. Recommended ≤0.125 µg/test (in 100 µL final volume). Cell number range: 10⁵–10⁸ cells/test. Optimize by titration for best performance.

CD25 is part of the IL-2 receptor complex regulating proliferation of T and B lymphocytes. The receptor is a heterotrimeric complex (IL2RA, IL2RB, IL2RG) forming high-affinity IL-2 binding sites. IL2RA homodimers form low-affinity receptors, while IL2RB homodimers form medium-affinity receptors. IL2 expression in mature thymocytes is monoallelic, ensuring precise regulation. IL-2 acts as a growth and differentiation factor and regulates apoptosis. It also stimulates B cell proliferation, enhances NK cell activity, and suppresses granulocyte-macrophage colony formation. IL2RA deficiency is associated with immune dysregulation disorders.
