Target Information
Mitochondrial transcription factor A (TFAM) is a crucial protein encoded by nuclear genes and transported from the cytoplasm to mitochondria, where it plays a key role in maintaining and expressing mitochondrial DNA (mtDNA). It regulates mtDNA replication and transcription, ensuring proper mitochondrial function.
Structurally, TFAM contains an LC3 interacting region (LIR) motif that binds to autophagy proteins such as LC3. This interaction supports the autolysosomal pathway, eliminating leaked mtDNA and limiting inflammation. TFAM levels vary across tissues, influencing mitochondrial function: high TFAM can repress mtDNA expression in skeletal muscle, while in cardiac tissue, balanced TFAM-to-mtDNA ratios preserve oxidative phosphorylation (OXPHOS) capacity.
Disruptions in TFAM can lead to mitochondrial dysfunction and are implicated in neurodegeneration and diseases related to mtDNA stress.
