Product Specific Information

The 93 monoclonal antibody reacts with an epitope shared by mouse CD16 and CD32.
CD16 (Fc gamma III Receptor) and CD32 (Fc gamma II Receptor) are low affinity receptors for the mouse IgG Fc portion and are expressed by B cells, monocyte/macrophages, NK cells, and neutrophils.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Tested by flow cytometric analysis of mouse splenocytes
Recommended usage ≤ 1.0 µg/test (100 µL final volume, 10⁵–10⁸ cells/test).
Careful titration is recommended for optimal performance.

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) must be used for optimal staining.
  • Add 5 µL per sample to antibody cocktails before combining with cells.
  • For single-color controls, use 5 µL per 100 µL cell sample containing 10³–10⁸ cells.

Target Information

CD16 is a 50–65 kDa surface molecule found in two forms:

• Transmembrane form (NK cells, some T cells)
• Phosphatidylinositol-linked form (granulocytes)

Functions as a low-affinity IgG receptor (FcR III), mediating ADCC by NK cells.

CD32 is a 40 kDa glycoprotein (Fc gamma RII) involved in:

• Endocytosis
• Secretion activation
• Cytotoxicity
• Immunomodulation

Expressed by B cells, monocytes, granulocytes, and platelets.
Involved in immune complex phagocytosis and regulation of antibody production.
Variants may increase susceptibility to systemic lupus erythematosus (SLE).
