Product Specific Information
- For IHC (PFA fixed): Incubate antibody for 30 minutes at room temperature.
- For formalin-fixed tissues: Heat tissue sections in 10 mM Tris with 1 mM EDTA (pH 9.0) for 45 minutes at 95°C, followed by cooling at room temperature for 20 minutes.
Target Information
CD22 (BL-CAM) is a type I integral membrane glycoprotein (130–140 kDa) expressed in the cytoplasm and membrane of B-lymphocytes. It appears early in B-cell differentiation, similar to CD19, and is expressed in mature B cells but not plasma cells. CD22 is strongly expressed in hairy cell leukemia and can be useful in phenotyping mature leukemias. It binds preferentially to α2,6-linked sialic acid and participates in regulation of B-cell receptor signaling through interactions with Src family kinases and cytoplasmic phosphatases. CD22 is positive in diffuse large B-cell lymphoma and nodular lymphocyte predominance Hodgkin’s lymphoma, but negative in classical Hodgkin’s lymphoma.
