• For IHC with PFA-fixed tissues: incubate antibody for 30 minutes at room temperature.
• Staining of formalin-fixed tissues requires heating in 10 mM Tris with 1 mM EDTA (pH 9.0) at 95°C for 45 minutes, followed by cooling at room temperature for 20 minutes.

Target Information

CD22 (BL-CAM) is a type I integral membrane glycoprotein with a molecular weight of 130–140 kDa. It is expressed in both the cytoplasm and cell membrane of B-lymphocytes and appears early in B-cell differentiation, similar to CD19.
Membrane expression is limited to mature B cells (CD22+) and absent in plasma cells (CD22–), making it useful for phenotyping mature leukemias. CD22 is strongly expressed in hairy cell leukemia and is positive in diffuse large B-cell lymphoma and nodular lymphocyte predominance Hodgkin’s lymphoma but negative in classical Hodgkin’s lymphoma.
It preferentially binds to α2,6-linked sialic acid, and its recognition site can be masked by cis interactions. Upon ligand-induced tyrosine phosphorylation, CD22 regulates B-cell receptor signaling by interacting with Src family kinases and recruiting phosphatases that modulate signaling pathways.
