Product Specific Information

The eBioCB16 monoclonal antibody recognizes CD16 (Fc gammaRIII), a low-affinity receptor for IgG with an apparent molecular weight of 50–80 kDa.
CD16 includes two genes:

• CD16A (FcγRIIIA) – membrane-anchored form in macrophages and NK cells
• CD16B (FcγRIIIB) – GPI-anchored form in neutrophils

Two polymorphisms (NA-1, NA-2) exist for CD16B; NA-2 homozygotes show lower phagocytic capacity.
CD16 binds IgG immune complexes, preferentially IgG1 and IgG3, and triggers ADCC, phagocytosis, degranulation, and proliferation upon activation.

Applications Tested: Pre-titrated for flow cytometry of normal human peripheral blood cells.
Use at 5 µL (0.06 µg) per test in a final volume of 100 µL (10⁵–10⁸ cells/test).
Excitation: 633–647 nm; Emission: 660 nm; Laser: Red Laser.
Filtration: 0.2 µm post-manufacturing filtered.

Target Information

CD16 encodes a receptor that recognizes the Fc portion of IgG, mediating clearance of immune complexes and antibody-dependent cytotoxicity.
The FCGR3A gene (CD16A) and FCGR3B gene (CD16B) are located on chromosome 1 and encode receptors expressed on NK cells and neutrophils, respectively.
Mutations in FCGR3A are associated with Immunodeficiency 20, Herpes Zoster, and susceptibility to systemic lupus erythematosus and neonatal neutropenia.
Alternative splicing produces multiple isoforms.
