Product Specific Information

The eBio500A2 monoclonal antibody reacts with the 25 kD epsilon subunit of the mouse CD3 complex. CD3 subunits gamma, delta, and epsilon are required for proper assembly, trafficking, and surface expression of the TCR complex. CD3 is expressed by thymocytes in a developmentally regulated manner and by all mature T cells and NKT cells.

Binding of eBio500A2 to CD3 initiates intracellular biochemical pathways that result in activation and proliferation. The 500A2 antibody partially cross-blocks the 17A2 and 145-2C11 antibodies, indicating recognition of distinct but overlapping epitopes.

  • Applications Tested: Tested on mouse splenocytes at ≤0.25 µg/test (100 µL sample volume, 10⁵–10⁸ cells/test).
    Optimal dilution should be determined empirically.

Fluorochrome Information:
APC-eFluor® 780 emits at 780 nm and is excited with the Red laser (633 nm). Ensure the instrument supports this fluorochrome.

Light Sensitivity:
This tandem dye is sensitive to photo-induced oxidation. Protect from light during storage and staining.

Fixation:
Samples can be stored in IC Fixation Buffer (Product #00-8222) or 1-step Fix/Lyse Solution (Product #00-5333) for up to 3 days at 4°C in the dark with minimal impact on brightness and FRET efficiency. Clone-specific performance should be verified empirically.

Target Information

The CD3 subunit complex is essential for transducing antigen-recognition signals into T cells and regulating TCR surface expression. Activation through TCR involves the cytoplasmic tails of CD3 gamma, delta, epsilon, and zeta subunits. These subunits are part of the immunoglobulin superfamily encoded on chromosome 11.

CD3 components interact with cytoplasmic signaling molecules via tyrosine-based motifs, contributing to TCR-induced growth arrest, survival, and proliferation. CD3 antigen is expressed on 68–82% of normal peripheral blood lymphocytes, 65–85% of thymocytes, and Purkinje cells, but not on B or NK cells. Altered T lymphocyte percentages may be observed in autoimmune diseases. Defects in CD3 genes are associated with CD3 immunodeficiency.
