Product Specific Information

The dG9 (delta G9) antibody reacts with human perforin (pore-forming protein, pfp), a cytolytic mediator found in cytotoxic T lymphocytes (CTLs) and natural killer (NK) cells. Perforin plays a critical role in immune defense against tumors and viral infections.

• Applications Reported: Intracellular staining followed by flow cytometric analysis.
• Applications Tested: Pre-titrated and validated for intracellular staining of normal human peripheral blood cells.
  • Use 5 µL (0.03 µg) per test (100 µL final volume).
  • Recommended cell range: 10⁵–10⁸ cells/test.

Fluorochrome Information:

• PerCP-eFluor® 710 can substitute for PE-Cy5, PE-Cy5.5, or PerCP-Cy5.5.
• Excitation: 488 nm (Blue laser)
• Emission: 710 nm
• Recommended filters: 685 LP dichroic mirror and 710/40 band pass filter (695/40 acceptable alternative).

Fixation Stability:
PerCP-eFluor® 710 conjugated antibodies remain stable when stained samples are fixed with 2% formaldehyde overnight at 4°C. Evaluate alternative fixation protocols as needed.

Filtration: 0.2 µm post-manufacturing filtered.

Target Information

Perforin is a major cytolytic protein stored in cytoplasmic granules of cytotoxic lymphocytes. It mediates pore formation in target cell membranes, facilitating immune defense against tumors and viral infections.

• Protein size: 555 amino acids (70–75 kDa)
• Structure: Contains a 21 amino acid signal peptide; homologous to complement component C9.
• Expression: Restricted to killer cell lines, not found in helper T cells or other tumor cells.
• Clinical relevance: Mutations in the perforin gene cause familial hemophagocytic lymphohistiocytosis type 2 (HPLH2).
• Alternative splicing: Multiple transcript variants reported.
