Cellular Localization Apical cell membrane; Single-pass type I membrane protein. Located in the apical domain of polarized epithelial cells. After endocytosis, internalized and recycled to the cell membrane. Found in microvilli and tips of filopodial protrusions. Isoform 5, Y, and 9 are secreted forms. Mucin-1 subunit beta localizes to cell membrane, cytoplasm, and nucleus. Upon EGF or PDGFRB stimulation, translocates to the nucleus via CTNNB1 interaction, enhanced by phosphorylation. Colocalizes with JUP/gamma-catenin in the nucleus upon HRG stimulation.

Background

This gene encodes a membrane-bound protein belonging to the mucin family. Mucins are O-glycosylated proteins essential for forming protective mucous barriers on epithelial surfaces and play roles in intracellular signaling. MUC1 is expressed on the apical surface of epithelial cells in tissues such as lung, breast, stomach, and pancreas. The protein undergoes proteolytic cleavage into alpha and beta subunits forming a heterodimeric complex. The N-terminal alpha subunit functions in cell adhesion, while the C-terminal beta subunit is involved in cell signaling. Overexpression, mislocalization, and altered glycosylation of MUC1 are associated with carcinomas. The gene contains a highly polymorphic VNTR domain, and alternative splicing results in multiple transcript variants.
