Product Specific Information

This 7G3 monoclonal antibody reacts with human and non-human primate CD123, the alpha chain of the IL-3 receptor. CD123 (60–70 kDa) binds IL-3 with low affinity alone, and with CD131 forms a high-affinity receptor complex.
CD123 is expressed by myeloid precursors, macrophages, dendritic cells, mast cells, basophils, and megakaryocytes, and is detectable on CD34⁺CD38⁻ hematopoietic stem cells and AML cells.

The 7G3 antibody inhibits IL-3-mediated proliferation and AML stem cell homing. Crossblocking studies show 7G3 recognizes a distinct epitope from 6H6.

Crossreactivity has been reported with chimpanzee, cynomologous monkey, olive baboon, pigtailed macaque, rhesus macaque, and sooty mangabey.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated for flow cytometric analysis of normal human peripheral blood cells.
Use at 5 µL (0.5 µg) per test (100 µL final volume). Cell number may range from 10⁵–10⁸ cells/test.

Fluorophore Information:
eFluor® 450 is an alternative to Pacific Blue®. Emits at 445 nm and excited at 405 nm (Violet laser). Ensure instrument compatibility.

Filtration: 0.2 µm post-manufacturing filtered.

Target Information

IL-3Ra (Interleukin-3 receptor subunit alpha) is part of the type I cytokine receptor family, forming a heterodimer with the common beta subunit (CDw131). IL-3Ra mediates IL-3 signal transduction.
Overexpression of IL-3Ra is observed on AML blasts compared to normal hematopoietic cells, making it a potential therapeutic target.
