Product Specific Information
The A7R34 monoclonal antibody reacts with mouse CD127, the high-affinity alpha subunit of the mouse IL-7 receptor. CD127 is expressed by immature B cells, double-negative (CD4-CD8-), single-positive (CD4+ or CD8+) thymocytes, but not double-positive (CD4+CD8+). In mature T cells, CD127 expression is low.
A7R34 inhibits IL-7 binding to its receptor and is used in studies of IL-7 roles in T/B cell development and activation. It blocks SB/199 antibody binding to mouse CD127.
Applications Reported: Flow cytometric analysis.
Applications Tested: Flow cytometric analysis of mouse splenocytes (≤1 µg/test).
A test is defined as staining a cell sample in 100 µL volume. Cell count typically ranges from 10⁵–10⁸ cells/test. Optimize antibody titration for best performance.
eFluor® 450 is an alternative to Pacific Blue®, emitting at 445 nm when excited at 405 nm (Violet laser). Ensure instrument compatibility.

Target Information
CD127 (Interleukin-7 receptor alpha) is a glycoprotein regulating lymphopoiesis. The IL7R complex consists of alpha and gamma chains; the gamma chain activates signal transduction, while the alpha chain mediates specific signaling.
CD127 promotes proliferation of precursor B cells, thymocytes, and mature T cells. Dysfunction is associated with severe combined immunodeficiency (SCID) variants.
