Product Specific Information

The 53-6.7 monoclonal antibody reacts with the mouse CD8a molecule, a 32–34 kDa cell surface receptor expressed as CD8αβ or CD8αα dimers. CD8 binds to MHC class I and, through association with p56lck, plays a role in T cell development and activation.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Mouse splenocytes at ≤0.5 µg/test (100 µL final volume; 10⁵–10⁸ cells/test)

Important Notes:

• NovaFluor dyes are incompatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assays.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
  • Add 5 µL per sample for antibody cocktails or single-color controls.

Target Information

CD8 (Cluster of Differentiation 8) is a type I transmembrane glycoprotein functioning as a co-receptor for MHC class I molecules. It is predominantly expressed on cytotoxic T lymphocytes (CTLs) and subsets of NK cells, monocytes, and dendritic cells. CD8 engagement triggers Lck kinase recruitment, leading to T cell activation and cytotoxic responses.

CD8 exists as:

• Heterodimer (CD8αβ) — on most T cells
• Homodimer (CD8αα) — on subsets of IELs and dendritic cells

Differentiation of naive CD8⁺ T cells into CTLs is enhanced by IL-2, IL-12, and TGF-β1.
