Target Information

Dynamin is a GTPase required for endocytosis of clathrin-coated vesicles from the membrane. Its role was first discovered in Drosophila melanogaster through the temperature-sensitive shibire mutation.
Three isoforms exist in mammals: Dynamin1 (neurons), Dynamin2 (ubiquitous), and Dynamin3 (testes).
Each isoform has multiple splice variants, suggesting distinct intracellular functions.
Common domain features include:

  • N-terminal GTPase domain
  • Central pleckstrin homology domain
  • Highly basic proline-rich C-terminal region with Src homology 3 (SH3) domains

These SH3 domains target dynamin to clathrin-coated pits. Dynamin interacts with AP2 adaptor protein complex and amphiphysin.
