Target Information
Leukocytes adhere to the blood vessel endothelium during extravasation in postcapillary venules of lymph nodes. Additionally, leukocyte adhesion occurs in capillaries and small venules throughout the body following inflammation. This rapid response involves a cascade of adhesion receptors. At the endothelial surface, members of the selectin and immunoglobulin superfamilies participate in this cascade.
E-selectin (CD62E) is an endothelial cell-specific selectin expressed only after activation by proinflammatory cytokines. In vitro, IL-1, TNF-α, and bacterial wall components such as lipopolysaccharides induce E-selectin transcription via an NF-κB dependent signaling cascade. E-selectin has been associated with vascular endothelium in various inflammatory conditions.
