Product Specific Information

Description:
The 2D1 monoclonal antibody reacts with all isoforms of human CD45 (Leukocyte Common Antigen, LCA). CD45 is expressed by all hematopoietic cells except circulating erythrocytes and platelets. The cytoplasmic portion of CD45 has tyrosine phosphatase activity and plays a key role in lymphocyte activation.

Each product contains one vial of NovaFluor conjugate and one vial of CellBlox Plus Blocking Buffer.

Applications Reported:
This 2D1 antibody is reported for use in flow cytometric analysis.

Applications Tested:
Pre-diluted and tested by flow cytometry using normal human peripheral blood cells. Recommended use: 5 µL (0.8 µg) per test in a final volume of 100 µL. Suitable for 10⁵–10⁸ cells/test.

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability analysis.
• Use CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) for optimal staining.
  • Add 5 µL per sample prior to mixing with cells.
  • For single-color controls, use 5 µL per 100 µL of cell sample (10³–10⁸ cells).

NovaFluor conjugates are based on Phiton™ technology using engineered nucleic acid dye structures designed to minimize spillover and spread in multicolor flow cytometry.

Target Information

CD45 (Leukocyte Common Antigen, LCA) is a receptor-type protein tyrosine phosphatase expressed on all nucleated hematopoietic cells, comprising about 10% of lymphocyte surface proteins. It is absent on non-hematopoietic cells and tissues. CD45 plays a critical role in lymphocyte development and antigen receptor signaling by regulating Src-family kinases and JAK kinases, influencing cytokine signaling, apoptosis, and integrin-mediated pathways. Multiple isoforms exist due to alternative splicing, differing mainly in extracellular domains.
