Product Specific Information

The monoclonal antibody Moushi101 recognizes mouse CD101, also known as IGSF2 and V7.
CD101 is a transmembrane glycoprotein with seven Ig-like domains and a short cytoplasmic tail containing phosphorylation sites.
Cross-linking CD101 can antagonize TCR/CD3-mediated lymphocyte activation.

Expression:
CD101 is expressed on monocytes, granulocytes, dendritic cells, Langerhans cells, and activated T cells.
It is also found on a subset of CD25⁺Foxp3⁺ regulatory T cells with higher suppressive properties in vitro and in vivo.

Included Components:

Applications Reported:
Suitable for flow cytometric analysis.

Applications Tested:
Tested on mouse splenocytes. Recommended ≤0.25 µg/test (100 µL final volume).
Cell number range: 10⁵–10⁸ cells/test.
Antibody should be titrated for optimal performance.

Compatibility Notice:
NovaFluor dyes are not compatible with DNA intercalating viability dyes such as:

  • Propidium iodide
  • 7-AAD
  • DAPI

Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.

CellBlox Plus Blocking Buffer:
Updated conjugates ship with CellBlox Plus Blocking Buffer (Cat. No. C001T06F01).
Add 5 µL per sample to antibody cocktails or single-color controls for optimal staining.

Technology:
NovaFluor conjugates utilize Phiton™ technology, based on novel nucleic acid dye structures engineered to minimize spillover and spread.

Target Information

CD101 (V7 antigen) is a member of the Ig superfamily and forms a disulfide-linked homodimer.
Expression is found on monocytes, granulocytes, dendritic cells (Langerhans-like), and T lymphocytes.
CD101⁺CD28⁺ cells respond strongly to CD28 signaling and can enhance proliferation with anti-CD28 or anti-CD3 stimulation.
Mouse CD101 is present on CD4⁺CD25⁺Foxp3⁺ regulatory T cells with high suppressor activity.
In humans, CD101 expression is observed on ~30% of Foxp3⁺ PBMCs, though suppressor activity is not confirmed.
