Product Specific Information

Description:
The monoclonal antibody 1A8-Ly6g reacts with mouse Ly-6G (Gr-1), a 25-kDa GPI protein expressed exclusively by neutrophils. It plays a role in neutrophil infiltration, recruitment, and migration, and associates with B2 integrins CD11a and CD11b. The ligand for Ly-6G remains unknown.

Unlike the RB6-8C5 antibody, 1A8-Ly6g binds only to Ly-6G on neutrophils and has been used for neutrophil depletion with reported antagonist activity.

Applications Reported:
Flow cytometric analysis.

Applications Tested:
Tested by flow cytometric analysis of mouse bone marrow cells. Recommended ≤1.0 µg per test (100 µL final volume). Optimize antibody titration empirically for best results.

Fluorochrome Information:
Brilliant Violet™ 650 (BV650) emits at 649 nm and is suitable for cytometers equipped with a violet (405 nm) laser. Ensure instrument compatibility before use.

Buffer Recommendations:
When using multiple polymer dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Cat. # SB-4401-42) or Brilliant Stain Buffer™ (Cat. # 00-4409-75) to minimize non-specific interactions.

Light Sensitivity:
Protect from light to prevent photo-induced oxidation.

Fixation:
Samples can be fixed with IC Fixation Buffer (Cat. # 00-8222-49) or 1-step Fix/Lyse Solution (Cat. # 00-5333-54) and stored up to 3 days at 4°C in the dark. BV650 is not compatible with methanol-based fixation.

Target Information

Ly6G (Lymphocyte antigen 6 complex locus G6D):
A component of the myeloid differentiation antigen Gr-1, expressed by myeloid-derived cells in bone marrow (21–25 kDa). Ly6G is a marker for neutrophils, monocytes, and granulocytes. Expression correlates with granulocyte differentiation and maturation. It may play a role in anti-tumor responses and modulation of local immune activity.
