Product Specific Information
The J105 monoclonal antibody reacts with human PD-1 (programmed death-1), a 55 kDa member of the CD28 immunoglobulin superfamily. PD-1 contains an immunoreceptor tyrosine-based inhibitory motif (ITIM) and plays a key role in peripheral tolerance and autoimmune disease. PD-1 is expressed predominantly on activated T and B lymphocytes. Its ligands PD-L1 (B7-H1) and PD-L2 (B7-DC) show overlapping functions and are expressed on normal tissues and activated antigen-presenting cells.
Costaining experiments suggest that eBioJ105 recognizes a different epitope than MIH4 (Product # 11-9969).
Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and tested by flow cytometry using normal human peripheral blood cells. Use 5 µL (1.0 µg) per test in a final volume of 100 µL. Cell number: 10⁵–10⁸ cells/test.
Fluorochrome Information:
Brilliant Violet™ 421 emits at 423 nm and is suitable for cytometers with a 405 nm violet laser.
When using multiple polymer dye-conjugated antibodies (e.g., Super Bright, Brilliant Violet™, Brilliant Ultra Violet™), use Super Bright Complete Staining Buffer (Product # SB-4401-42) or Brilliant Stain Buffer™ (Product # 00-4409-75) to minimize non-specific interactions.

Target Information
T lymphocytes initiate cell-mediated immune responses upon stimulation by peptides presented on MHC molecules of antigen-presenting cells (APCs). Activation is self-limited as PD-1 (PDCD-1) mediates inhibitory signaling upon binding with its ligands PD-L1 or PD-L2. These interactions suppress immune activation in the absence of danger signals. PD-1-deficient mice exhibit hyperactivated immune systems and autoimmune diseases. PD-1 often migrates at a higher molecular weight than predicted in SDS-PAGE.
