Product Specific Information
The 145-2C11 monoclonal antibody reacts with mouse CD3e, a 20 kDa subunit of the TCR complex. Along with other CD3 subunits (gamma and delta), the epsilon chain is required for proper assembly, trafficking, and surface expression of the TCR complex. CD3 is expressed by thymocytes and all mature T cells. Binding of 145-2C11 to TCR initiates intracellular signaling leading to activation, proliferation, or apoptosis depending on conditions.
Applications Reported: Flow cytometric analysis
Applications Tested: Tested on mouse splenocytes at ≤1.0 µg/test (100 µL final volume). Optimal titration recommended.
Fluorophore Information:
Brilliant Violet™ 421 (BV421) emits at 423 nm; suitable for violet (405 nm) laser-equipped cytometers.
When using multiple polymer dye-conjugated antibodies (Super Bright, Brilliant Violet™, Brilliant Ultra Violet™), use Super Bright Complete Staining Buffer (Product # SB-4401-42) or Brilliant Stain Buffer™ (Product # 00-4409-75) to minimize non-specific interactions.

Target Information
The CD3 subunit complex is essential for transducing antigen-recognition signals into T cells and regulating TCR surface expression. CD3 subunits (gamma, delta, epsilon, zeta) are members of the immunoglobulin superfamily encoded on chromosome 11. They possess cytoplasmic tails associated with signal transduction molecules via tyrosine-based motifs.
CD3 is expressed on:
- 68–82% of normal peripheral blood lymphocytes
- 65–85% of thymocytes
- Purkinje cells in the cerebellum
Not expressed on B or NK cells. Altered T lymphocyte levels may occur in autoimmune diseases. Defects in CD3 gene are linked to CD3 immunodeficiency.
