Product Specific Information
The eBioRDR5 monoclonal antibody reacts with human CD127 (Interleukin-7 Receptor alpha). CD127 complexes with CD132 (common gamma chain, γc) to form the IL-7 receptor (IL-7R).
CD127 is a type I glycoprotein (75–80 kDa) expressed by immature B cells, thymocytes, and most mature T cells, with transient down-regulation upon activation.
IL-7 binding triggers signal transduction via tyrosine kinase pathways, including Jak/STAT. IL-7 is essential for lymphocyte development and homeostatic proliferation of T cells.
IL-7R signaling also regulates TCR locus rearrangement in γδ T cells.
CD127 expression is down-regulated on CD4⁺CD25⁺ regulatory T cells (Tregs). Cells with low CD127 and high CD25 are enriched for regulatory T cells expressing Foxp3 and suppressive activity.
Binding of eBioRDR5 to PBMCs is blocked by pre-incubation with recombinant human IL-7 (Product #14-1079-80).

Applications Tested: Pre-titrated for flow cytometry using normal human peripheral blood cells at 5 µL (0.06 µg)/test (100 µL final volume).

eFluor® 660 is a replacement for Alexa Fluor® 647, excited by red laser (633 nm) and emitting at 659 nm.
Target Information
CD127 (Interleukin-7 Receptor alpha, IL7Rα) is a glycoprotein regulating lymphopoiesis. The functional receptor is an α/γ heterodimer (IL7Rα and γc).
The γc chain activates signal transduction, while the α chain mediates specific signaling events.
CD127 promotes proliferation of precursor B cells, thymocytes, and T cell subsets.
Dysfunction is associated with severe combined immunodeficiency (SCID) and related disorders.
